以枸杞品种蒙杞1号和宁杞1号为材料,通过制浆、盐析、透析、浓缩、电泳、分析等方法对其中的蛋白质进行提取分离、鉴定,探讨其在不同pH和(NH4)2SO4浓度梯度下蛋白质的差异。结果表明,蒙杞1号在相同pH下,(NH4)2SO4浓度为40%时蛋白质含量最多,共有7种亚基, (NH4)2SO4浓度为80%时基本没有蛋白质;在相同(NH4)2SO4浓度下,pH 6时亚基数为7,即蒙杞1号在pH 6.0、 (NH4)2SO4浓度40%时提取效果最佳。宁杞1号在相同pH下, (NH4)2SO4浓度为60%时蛋白质含量最多,共有5个亚基;在相同(NH4)2SO4浓度下,pH 7.0时亚基数为5,即宁杞1号在pH 7.0、 (NH4)2SO4浓度为60%时提取效果最好。

Mengqi 1 and Ningqi 1 Lycium barbarum L. were used as materials to extract and identify proteins by pulping, salting out, dialysis, concentration, electrophoresis, analysis and other methods, so as to explore the differences of proteins under different pH and(NH4)2SO4 concentration gradient. The results showed that Mengqi 1 had the most protein content when(NH4)2SO4 concentration was 40% at the same pH value, with a total of 7 subunits. When(NH4)2SO4 concentration was 80%, there was basically no protein; under the same concentration of (NH4)2SO4, the subunits is 7 at pH 6.0, that is, Mengqi 1 has the best extraction effect at pH 6.0 and(NH4)2SO4 concentration at 40%. Ningqi 1 had the most protein content when (NH4)2SO4 concentration was 60% at the same pH value, with 5 subunits in total; under the same(NH4)2SO4 concentration, the subunits is 5 at pH 7.0, that is, Ningqi 1 has the best extraction effect when pH 7.0 and (NH4)2SO4 concentration is 60%.
